The parasite-specific trypanothione metabolism

چکیده

Trypanosomes and leishmania are the causative agents of African sleeping sickness (Trypanosoma brucei), South American Chagas’ disease (T. cruzi) and other tropical diseases. All these parasitic protozoa have a unique thiol metabolism. Instead of the ubiquitous glutathione, the main non-protein thiol is trypanothione [bis(glutathionyl)spermidine] which is kept reduced by the flavoenzyme trypanothione reductase (Figure 1). Aim of our work is to elucidate the specific functions of the trypanothione metabolism and to contribute to the development of new antiparasitic drugs on the basis of specific enzyme inhibitors.

The parasite-specific trypanothione redox metabolism

Research Highlights Dissecting the mechanism of trypanothione synthetase under in vivo-like conditions All proteins of the trypanothione metabolism are indispensable for the viability of African trypanosomes. A final conclusion as to which protein plays a main role in the pathway control yet would need a reliable model based on the kinetic parameters of all enzymes obtained under conditions tha...

Major therapeutic targets in trypanosomatids

Trypanosomatids are protozoan parasites that have some peculiar characteristics in biological terms, such as the presence of a single flagellum and kinetoplast (i.e., a specialized organelle that concentrates mitochondrial DNA in the parasite) and alternating forms in vectors and hosts with distinct modes of morphology, biochemistry, and exhaust. Among the diseases caused by trypanosomatids are...

Thiol-based redox metabolism of protozoan parasites.

The review considers redox enzymes of Plasmodium spp., Trypanosomatida, Trichomonas, Entamoeba and Giardia, with special emphasis on their potential use as targets for drug development. Thiol-based redox systems play pivotal roles in the success and survival of these parasitic protozoa. The synthesis of cysteine, the key molecule of any thiol metabolism, has been elucidated in trypanosomatids a...

Purification and characterization of a trypanothione-glutathione thioltransferase from Trypanosoma cruzi.

Although trypanothione [T(S)2] is the major thiol component in trypanosomatidae, significant amounts of glutathione are present in Trypanosoma cruzi. This could be explained by the existence of enzymes using glutathione or both glutathione and T(S)2 as cofactors. To assess these hypotheses, a cytosolic fraction of T. cruzi epimastigotes was subjected to affinity chromatography columns using as ...

Identification of Novel Chemical Scaffolds Inhibiting Trypanothione Synthetase from Pathogenic Trypanosomatids

BACKGROUND The search for novel chemical entities targeting essential and parasite-specific pathways is considered a priority for neglected diseases such as trypanosomiasis and leishmaniasis. The thiol-dependent redox metabolism of trypanosomatids relies on bis-glutathionylspermidine [trypanothione, T(SH)2], a low molecular mass cosubstrate absent in the host. In pathogenic trypanosomatids, a s...
